Charlotte was born as Sophia Charlotte, the youngest daughter of Duke Charles Louis Frederick of Mecklenburg-Strelitz and Princess Elizabeth Albertina of Saxe-Hildburghausen, at Untere Schloss (Lower Castle) in Mirow.

Her brother Adolphus Frederick succeeded to the ducal throne, giving Charlotte her first experience of princely duties and court life.
